PLANT DNA AND TISSUE PREPARATION

The University of Florida Herbarium (FLAS) of the Florida Museum of Natural History and the Soltis Lab conduct an array of projects with many plant groups.
The GRR generally processes plant material obtained from the collecting trips of their associated researchers.
In the field, herbarium and tissue specimens are normally prepared and, depending on the project, anatomical samples and root tips for chromosome count are also prepared. The GRR stores tissue and DNA samples obtained from herbarium sheets or tissues that have been dried and stored in silica gel. DNA samples are used in phylogenetic and population studies and the remaining DNA is stored in the GRR. We try to store at least 50ul of total DNA per sample with documentation of its concentration and collecting information (locality, date and collector, collector number, etc).
